DOES ZYMŌT SPERM SEPARATION IMPROVE EMBRYO DEVELOPMENT OUTCOMES WHEN APPLIED TO ALL INFERTILITY PATIENTS COMPARED TO DENSITY GRADIENT WASHING OR SURGICALLY ATTAINED SPERM?

چکیده
The ZyMōt sperm separation device has proven favorable for use in elevated DNA fragmentation index (DFI) male factor patients, as an alternative to density gradient (DG) washing or surgically attained testicular sperm. In 2020, without fully understanding the infectivity and transmission potential of SARS-CoV2 semen, a more liberal application timed microfluidic swim-up was applied our IVF patients dilute out minimize pathogens. This study aimed evaluate whether improved normal embryo development. Retrospective analysis PGT-A/ICSI cycles (N= 3219) between 2016-2020 conducted assessed fertilization rates (FR), blastocyst development/utilization (BUR) genetic outcomes. Sperm preparations were performed per standard operating manufacturer advised (i.e., ZyMōt) procedures. Cumulus oocyte complexes harvested 36h post-hCG, stripped ICSI 3-5hr later. Zygotes at 16-18hr post-ICSI, embryos cultured under humidified tri-gas incubation up 7 days. Blastocyst (BL) development evaluated, expanded BL greater biopsied on Days 5, 6 7. All vitrified genetics determinations euploidy, aneuploidy mosaicism contrast. Applying Chi-squared analysis, we compared differences (p<0.05) oocytes inseminated by DG wash (n=23,549), (n=7,331) (n=815). No difference FR (76%), D5 formation (52-56%) BUR (52-53%) detected washed sperm, respectively. Meanwhile, had lower (70%; p<0.05), fewer forming (48%; p<0.05) overall (41%; p<0.05). addition, testicular-derived euploid (39%; with (54%; than (50%, 39%; respectively) (45%, 37%; derived-embryos. potentially viable (Euploidy+Mosiac outcome) observed groups (63-64%). Application general population offered no benefit outcomes Our data does support that using DFI is cost-effective approach attaining when ejaculated possible. However, insufficient motile morphologically are available ejaculate further needed elucidate biopsy treatment, assessments this may be biased including men non-obstructive azoospermia.

Effectiveness of sperm washing by discontinuous density gradient centrifugation to remove antibodies bound to the sperm membrane
OBJECTIVE:We evaluated the effectiveness of spermwashing using the discontinuous two-layer density gradient centrifugation method to remove antisperm antibodies attached to the sperm surface. METHOD: We prospectively enrolled sixty-six men with unexplained infertility who were seeking evaluation. Each patient delivered one semen specimen for the study. We determined antisperm antibody levels us...
متن کاملThyroxin Is Useful to Improve Sperm Motility
Objective The aim of this study was to evaluate the non-genomic action of thyroxin on sperm kinetic and its probable use to improve sperm recovery after applying an enrichment method like “swim-up” in comparison with the available one, pentoxifylline. MaterialsAndMethods This is an experimental study. A total of 50 patients were recruited, followed by infertility consultation. Conventional sper...
متن کاملcan zeta sperm selection method, recover sperm with higher dna integrity compare to density gradient centrifugation?
background: sperm selection for icsi based on morphology and motility might not be relevant to chromatin integrity. thus sperm selection based on sperm characteristics has been suggested. objective: the aim of this study was to compare the efficiency of zeta method with routine density gradient centrifugation method (dgc) for the selection of sperm with higher dna integrity. materials and metho...
متن کاملSperm Preparation Techniques for Artificial Insemination - Comparison of Sperm Washing, Swim Up, and Density Gradient Centrifugation Methods
The Artificial Insemination (AI) is the first option treatment for infertile couples with cervical factor subfertility, mild-moderate male subfertility and unexplained infertility. With the exception of cases in which the use of in vitro fertilization (IVF or ICSI) is strictly due as a consequence of a severe male or female factor, the artificial insemination must be part of a gradual approach ...
متن کاملComparison of Sperm Telomere Length between Two Sperm Selection Procedures: Density Gradient Centrifugation and Zeta Potential
Background: Telomeres are particular sequences of DNA located at the end of the eukaryotic chromosomes that are essential for genome integrity. Telomere length in spermatozoa differs among males, as well as spermatozoa. Also, decreased telomere length in spermatozoa of infertile men is associated with the reduction of fertility potential and embryo quality. Density gradient centrifugation (DGC)...
